preparation | 3-acetyl-7 (1H)-azaindole can be used as a pharmaceutical synthesis intermediate, it can be prepared by the reaction of 7-azaindole with acetyl chloride. Preparation of 3-acetyl-7 (1H)-azaindole: 7-azaindole (590mg,5mmol) was dissolved in 80ml DCM and AlCl3(3.39g,25mmol) was added, slowly add acetyl chloride (3.4mL,47.8mmol), react at room temperature for 10h, place the reaction solution in ice bath, slowly add methanol 20mL to quench the reaction, concentrate the reaction solution, add 30ml water to the residue, adjust PH ~ 4 with 1n NaOH, EtOAc(15ml × 5) extraction, anhydrous sodium sulfate drying, concentration, column chromatography separation (P:E = 1:1), 3-acetyl-7 (1H)-azaindol was obtained as a white solid in 82.5% yield. 1HNMR(400MHz,CDCl3)δ13.11-12.78(m,1H),8.79-8.69(m,1H),8.47-8.37(m,1H),8.15-7.98(m,1H),7.36-7.28(m,1H),2.64-2.52(m,3H). 13CNMR(100MHz,CDCl3)δ193.43,149.21,143.54,132.43,131.83,118.96,118.44,116.79,27.15. |
